Adenosine contributes to complement‐activation‐related paradoxical bradycardia

Abstract

Intravenous injection of complement (C)‐activating drugs in pigs can cause immediate cardiac anaphylaxis associated with myocardial ischemia and relative bradycardia during severe hypotension. We tested whether this paradoxical bradycardia could be mediated by myocardial adenosine. Pigs were injected i.v. with adenosine or various C‐activating drugs with or without pretreatment with nonselective adenosine receptor (A) blocker theophylline, or selective adenosine A1/A2 receptor antagonists cyclopentyl‐xanthine (CPX) and ZM24135, respectively. ECG and hemodynamic parameters were recorded.
