ORGANO-TIN TOXICOLOGY

Abstract

MITOCHONDRIAL SWELLING AS EVIDENCED BY A DECREASE IN OPTICAL DENSITYWAS ALWAYS OBTAINED WITH CONCENTRATIONS OF TRIETHYLTIN (TET) AS LOW AS 1 X 10-7M. THE SWELLING INDUCED BY TET WAS UNAFFECTED BY THE ADDITION OF ETHYLENEDIAMINE TETRACETATE (EDTA) INDICATING THAT THE SWELLING OBSERVED WAS NOT THE RESULT OF AN IMPURITY SUCH AS INORGANIC TIN. IT WAS ALSO DEMONSTRATED THAT TET INHIBITS THE ATP REVERSAL OF SWELLING INDUCED BY THYROXIN. TET-INDUCED SWELLING (10-7M TET) COULD BE REVERSED BY ATP ADDITION, LESS READILY BY ADP AND NOT AT ALL BY AMP. FURTHER, THE ADDITION OF MN ION TOGETHER WITH THE ATP SEEMED TO POTENTIATE THE REVERSAL. WITH THIS COMBINATION IT WAS POSSIBLE TO REVERSE TETINDUCED SWELLING WHEN TET WAS PRESENT IN CONCENTRATIONS AS HIGH AS 10-4M. WHEN 2.4 DINITROPHENOL (2.4 DNP) WAS ADDED TO THE MEDIUM PRIOR TO TET ADDITION, IT EFFECTIVELY PREVENTED THE USUAL SWELLING
