STUDY OF SECONDARY DETERIORATION IN PARTIALLY OXIDIZED LIPIDS

Abstract

GELATIN AND EGG ALBUMIN FOAMS CONTAINING METHYL LINOLEATE WERE PREPARED, AND AFTER AGING IN AIR WERE HYDROLYZED WITH TRYPSIN. THE PRESENCE OF AUTOXIDIZED METHYL LINOLEATE IN THE FOAM WAS FOUND TO SIGNIFICANTLYALTER THE HYDROLYTIC PROPERTIES OF TRYSIN TOWARD THE TEST PROTEINS.IN GELATIN IT WAS FOUND THAT 22( LESS N WAS LIBERATED IN A FAT OXIDIZED SAMPLE THAN IN A CONTROL FOAM. THIS RESULT IS INTERPRETED AS CONFIRMING EVIDENCE THAT PROTEINS IN INTIMATE CONTACT WITH METHYL LINOLEATE BECOMES CROSS-LINKED DURING AERATION OF THE MIXTURE. IN EGG ALBUMIN,ON THE OTHER HAND, THE PRESENCE OF AUTOXIDIZED METHYL LINOLEATE IN THE FOAM GREATLY ENHANCES THE HYDROLYTIC POWERS OF TRYPSIN. THIS APPARENT INCONSISTENCY IN THE ENZYME DATA MAY BE EXPLAINED AS BEING DUE TO THE PRESENCE OF A STRONG TRYSIN INHIBITOR, OVOMUCOID, WHICH IS A COMMON CONTAMINANT IN IMPURE EGG ALBUMIN. PRELIMINARY BIOASSAY STUDIES OF THE INSULIN-METHYL LINOLEATE INTERACTION HAVE BEEN CONCLUDED. AFTER 4 DAYS' REACTION WITH METHYL LINOLEATE (IN OXYGEN), THE POTENCY OF INSULIN AS EXPRESSED IN TERMS OF CONVULSIVE DOSE (CD) WAS REDUCEDFROM 90 TO 63(. AT THE END OF 29 DAYS, THE POTENCY WAS REDUCED FROM CD 83( (CONTROLS) TO O FOR SAMPLE CONTAINING METHYL LINOLEATE. THIS STUDY IS IMPORTANT FOR IT INDICATES THAT INSULIN BIO-ASSAY PROVIDES A SENSITIVE MEANS OF FOLLOWING FAT-PROTEIN INTERACTIONS.
