FURTHER STUDIES ON THE DISTRIBUTION AND METABOLISM OF 2-MERCAPTOETHYLGUANIDINE AND BIS (2-GUANIDOETHYL) DISULFIDE IN MICE

Abstract

GED = bis(2-guanidoethyl)disulfide. The distribution of S35 in the various tissues and the metabolism of 2-mercaptoethylguanidineS-35 (MEG-S35) and bis(2-guanidoethyl)disulfideS-35 (GED-S35) as determined by chromatography of tissue homogenates were studied in 8- to 10week-old C57BL/6J male mice. Protein-bound S35, GED, taurocyamine, guanidoethylsulfinic acid, MEG, and sulfate were identified in the tissues. Several unknown compounds were also observed. Concentrations of protein-bound S35 and GED decreased at 120 min after GED administration and correlated best with protection of the mice. All products increased in the tissues at the higher dose of MEG, but the protein-bound S35 did not increase as much as other products. Some differences in the concentrations of products were noted on compaison of MEG-trea ed with GED- treated animals. The relationship of these results to mechanism of action of MEG and GED is discssed. (Author)
