REACTIVITY OF SULFHYDRYL GROUPS IN BOVINE PLASMA ALBUMIN BEFORE AND AFTER ULTRAVIOLET IRRADIATION.

Abstract

A study of the activity of sulfhydryl and disulfide groups in bovine plasma albumin after various periods of ultraviolet irradiation has been performed. The reactivity of these groups was studied by way of amperometric titrations using mercuric chloride, ethylmercuric chloride (EMC) and silver nitrate as titrating agents. Upon irradiation, disulfide groups of albumin are split and the titratable amount of sulfhydryl increases. Under the experimental conditions the maximum number of disulfide groups which can be broken by UV irradiation was found to be eleven, while the remaining six disulfide groups remain unattacked by irradiation. This was found for albumin solutions irradiated in the absence and presence of 4F quanidine hydrochloride. Apparently the radiation effect for albumin, as far as the disulfide bonds are concerned, is the same, no matter whether the protein is initially in the native or unfolded state. It can be concluded that the changes in the secondary structure of albumin are of similar nature in guanidine and radiation denatured protein. (Author)
