A METHOD FOR THE ESTIMATION OF GELATIN IN THE PRESENCE OF SERUM PROTEINS AND NON-PROTEIN NITROGEN.

Abstract

This report describes a reliable method for the estimation of gelatin, based on a dilution principle, in which the composition of a given mixture may be obtained from a ratio of its chromogenic groups to its total nitrogen content. Since the composition ratios are based entirely on nitrogen rather than protein the results are independent of inequalities of protein-nitrogen conversion factors which may exist between the two components and may be reported in terms of gelatin-N concentrations. If the conversion factor for gelatin is given the former value can then be easily converted to yield the true concentrations of gelatin in the sample. Owing to the fact that the relationship between chromogenic and nitrogen values is linear for both gelatin and plasma protein and that the two lines originate from a determined common extrapolated X-intercept, it is necessary only to calculate the true slopes of these lines in a standardization experiment in order to construct a composition graph to which corrected unknown ratios may be referred.
