FRACTIONATION OF BACTERIOPHAGE SP-10 DNA ON HYDROXYAPATITE COLUMNS,

Abstract

Bacteriophage SP-10 DNA may be fractionated by column chromatography on hydroxyapatite. Two components are readily isolated. One component consists of low molecular weight fragments. This component exhibits a typical single-strand thermal denaturation profile. The second component appears to be native double-strand DNA and exhibits a thermal denaturation profile indicative of double-stranded DNA. The original DNA preparation (prior to chromatography) exhibits a thermal denaturation profile which is the sum of the two individual profiles obtained from the column fractionated components.
