A HISTIDINE ACTIVE-CENTER DECAPEPTIDE FROM CHYMOTRYPSIN.

Abstract

Chymotrypsin was inactivated with C14-labeled N-tosyl-L-phenyl-alanine chloromethyl ketone (TPCK), and the inactive enzyme was cleaved with performic acid and pepsin. A histidine active-center decapeptide was isolated by chromatography with Sephadex G-50, DEAE Sephadex A-25, Sephadex G-25, and Dowex 1-X2. Amino acid composition and end-group analysis showed its structure to be NH2-ala(ala, his-TPCK, CySO3H, thr, thr, gly, val, ser)-asp-COOH. (Author)
