A STANDARD TECHNIQUE FOR TEMPORAL BONE PREPARATION,

Abstract

The inner ear end organs are structurally fragile with the membraneous labyrinth weakly supported within the bony labyrinth. The technique of histological preparation must be directed toward preserving the anatomical relationship of the inner ear structures and minimizing post-mortem changes. The densely ossified petrous portion of the temporal bone must be decalcified and the inner ear spaces uniformly infiltrated with celloidin, all the while maintaining the histological integrity of the membranous labyrinth. This monograph is intended as a guide in the preparation of good temporal bone slides which will make it possilbe to investigate the correlation between end organ functions and morphological findings. No fundamental histological information is included, however, as that appears in other publications. The techniques described herein have been found to render the most consistently satisfactory temporal bone preparations. (Author)
