RAPID IDENTIFICATION OF MICROORGANISMS BY CONTINUOUS PARTICLE ELECTROPHORESIS.

Abstract

The feasibility of using continuous particle electrophoresis for fractionating and identifying certain specified biologic organisms was investigated. To determine the optimum operating conditions for achieving this goal, various procedures were used, including different bacterial handling methods, various pH values of the certain buffer, and various sterilization procedures. It was found that the five microorganisms of interest could be separated from each other by judicious choice of operating conditions. Several technics potentially capable of automatic screening of unknown bacterial suspensions were also studied. These studies used the Beckman optical scanner to detect microorganisms, determine their electrophoretic mobility, and determine the effect of pH gradients on electrophoretic mobility. These studies resulted in a procedure which may be feasible for completely automatic detection and characterization of microorganisms within a mixture, if the prior history of the mixture is known.
