FLUORESCENCE SPECTRA, POLARIZATION, AND LIFE TIMES OF BIOLOGICAL COMPOUNDS IN LIVING CELLS AND IN MODEL SYSTEMS.
Abstract
The overall purpose of the study is to produce data representing three major fluorescence parameters from biological compounds while these compounds are in interaction with living cells. The three parameters are fluorescence spectra, lifetime (or decay time), and polarization. This data is to be used in the interpretation of molecular binding effects such as energy transfer, degrees of freedom, macromolecular volumes, and binding strengths. In this annual report there appears: (A) Descriptions of optical bench construction for microscopic measurement of the three fluorescence parameters mentioned above. This bench includes an intensifier orthicon microspectrofluorimeter, a Pockels cell arrangement for decay time measurement, and polarizers for the fluorescence polarization data. (B) Pilot data on fluorescence decay times of a number of compounds complexed with cells in suspension. The compounds were chosen as representative of different types of binding, some involving energy transfer and others not. (Author)
