A BIPHASIC EXTRACTION FOR 2,8-DIOXYADENINE.

Abstract

A rapid quantitative method for 2,8-dioxyadenine in biological fluids has been developed using a biphasic HCl extraction from Dowex-50W, X-8, 200-400 mesh, hydrogen form. The precision and accuracy of this assay were evaluated. The least square fit of the standardization gives a slope of 0.999 plus or minus 0.0647 resulting in essentially 100% recoveries. When compared with the 2-day column chromatography technique previously available, this procedure permits numerous determinations within the hour. The result is a rapid screening assay for 2,8-dioxyadenine whenever large doses of adenine are administered. A variation of this procedure may be used as a semiquantitative test for adenine determination. (Author)
