Isolatio and Identification of 5-Methylthioribose in Escherichia coli B.

Abstract

5-Methylthioribose was isolated after incubation of Escherichia coli B in a glucose-salts medium. At least sixty per cent of the radioactivity in absolute ethanol extracts of the residue from lyophilized medium supplemented with 35SO4(2-) was located in two chromatographic areas that represented 5-methylthioribose and its sulfoxide, an artifact. These compounds were characterized by functional group analysis and chromatographic comparison with authentic material. Fifty micrograms of 5-methylthioribose sulfoxide were isolated from the incubation medium of E. coli and purified in four paper and thin layer chromatographic systems. The trimethylsilyl derivative of this compound was compared with the derivative of authentic 5-methylthioribose sulfoxide obtained following hydrolysis of 5'-methylthioadenosine isolated from Saccharomyces cerevisiae. Gas chromatography and mass spectrometry confirmed the identity. This is the first report of 5-methylthioribose in a bacterium. (Author)
