Some Metabolites of CS from Rats.

Abstract

The purpose of this study was to obtain qualitative and quantitive information about the major metabolites of o-chlorobenzalmalononitrile (CS). CS in PEG-200 solution was administered intragastrically to rats. Urine was collected from control and treated animals for 24 hours. The urinary acids were isolated and converted into their respective methyl esters for gas chromatographic analysis. Several of the esters eluted from the chromatographic column were passed into a mass spectrometer, and the urinary acids from the CS-treated rats were identified by mass spectrometry. The following were concluded. (1) A major metabolite of CS administered intragastrically to the rat is o-chlorohippuric acid. The amount recovered was 33% of the amount theoretically derivable from the weight of CS administered. Another metabolite is o-chlorobenzoic acid, of which 1% was recovered. (2) The identification of o-chlorohippuric acid as a major metabolite of CS strongly suggests that CS is hydrolyzed to o-chlorobenzaldehyde and malononitrile in the rat gut. The aldehyde is oxidized to o-chlorobenzoic acid, which then is conjugated with glycine to form o-chlorohippuric acid. (Author)
