Viability of Stored Tissue.
Abstract
The objective of this research is to develop cryoprotective procedures which will allow long-term organ storage and eventually lead to the transplantation of cadaver kidneys on a wide spread scale. In this regard, it is necessary to be able to evaulate the functional capabilities of the kidney prior to transplantation. Using the isolated perfused canine kidney as a model system, a series of physiological metabolic parameters have been developed which can be measured during in vitro perfusion and can provide an index of the viability of the organ. The experimental protocol for renal perfusion will be described and a series of typical results of a kidney perfused at 25C will be presented. In addition, in vitro experiments will be described which will enable the measurement of glycolysis and oxidation in renal cell suspensions under hypothermic conditions.
