The Endogenous Metabolism of Anaerobic Bacteria.

Abstract

An investigation of the possible role of the tricarboxylic acid cycle in the anaerobe Zymomonas anaerobia has been carried out. he enzymic constitution of cells grown in both complex and defined medium has been examined and only four enzymes of the cycle, namely citrate synthase, aconitase, isocitrate dehydrogenase and malate dehydrogenase detected. With the exception of malate dehydrogenase, the specific activities of these enzymes are similar to those reported by other workers for anaerobically-grown Escherichia coli. The complementary approach of paper, thin-layer and column chromatography of intermediates extracted from cells that have metabolized unlabelled or (U-14C) glucose permitted the detection of small amounts of all the intermediates of the cycle. Washed suspensions of the organism incorporate 14C from (U-14C) aspartate into all the intermediates of the cycle and also into some cellular components. The organism contains small amounts of haem and porphyrin and the problem of its biosynthesis during growth in defined medium is discussed. It did not prove possible to induce enzymes of the cycle in cells which were aerated in the complex growth medium. (Author)
