Structure-Function Analysis of Antimalarial Compounds.
Abstract
The object of the research was to subject the multitudinous antimalarial structure-function data available to careful examination, using the multiple parameter statistical analysis approach and thereby to obtain mathematical equations which relate physical-chemical properties (both measured and calculated) to biological test results. Then these results were to be compared with those obtained by application of the Free-Wilson technique. Comparison and study of these equations would assist attempts to separate toxicity from antimalarial activity, and would guide the development of the most effective antimalarial compounds possible in each chemical series thus examined. The report contains the following: Studies of the 8-aminoquinolines; Studies of the quinoline carbinols by the Free-Wilson technique; Studies of the phenanthrene carbinols by both the Hansch and Free-Wilson techniques; Further development of the Free-Wilson method.
