Studies of the Mammalian Brain Function in vitro.

Abstract

From the studies of the cat brain function it has been thought that the most important factors which decide the sustained reversibility of the function of a frozen brain depends on preservability of the vascular structure in the brain against freezing damage. In this experiment it was intended to compare the effects of three cryoprotective substances, i.e. dimetyl sulfoxide (DMSO), glycerol and polyvinyl pyrorridone (PVP) on the vascular structure of a brain after freezing and thawing. The brain perfused with dextran-Hanks solution in situ was isolated and cryoprotective agent was added in the perfusate. It was immersed in the fluid containing the same cryprotective agent, and frozen at -20C for one week. After thawed and expelling the cryoprotective substances, indian ink was added to the perfusate. To compare the effect of the cryoprotective agents, the three brains treated with DMSO, glycerol and PVP respectively and perfused at the same time and with the identical pulsatory perfusion pressure. The photographs were taken at intervals and the appearance of macrocerculation was compared. After elapse of 45 sec ink perfusion was stopped and histological sections were made from the individual brain. From the results obtained, DMSO was the most effective preservative for both preservation of capillary and electrical activity of the brain. (Author)
