Effect of Multiple Additions of Inosine and a Single Addition of Adenine on the 2,3-DPG, ATP and P sub 50 of Stored Red Cells, and the Additive Effect of Inosine and of Anticoagulant ACD pH 5.3 on the Maintenance and Regeneration of 2,3-DPG.

Abstract

Addition of a constant amount of adenine (2 micromoles/gm. of hemoglobin) and a variable amount of inosine have resulted consistently in a significant increase of the level of 2,3-DPG of red cells of ACD stored blood; the level of 2,3-DPG was found directly related to the concentration of inosine. Twice weekly additions of 10 micromoles of inosine for a total of 120 micromoles/gm. of hemoglobin to ACD-adenine blood resulted in the maintenance of 2,3-DPG level above 90% of the original for 42 days. Addition to fresh blood collected in ACD pH 5.3 of inosine only, in concentrations of 30 to 120 micromoles per gram of hemoglobin showed maintenance of 2,3-DPG at levels significantly higher than found in the fresh blood; the P50 was similarly preserved. With 60 micromoles of inosine/gm. of hemoglobin, 2,3-DPG was maintained at levels significantly higher than that of the fresh blood for 28 - 29 days. Results with addition of 30 micromoles of inosine were not as good, but increasing the dose above 60 micromoles did not improve the level of 2,3-DPG or P50. Addition at 14 days of 2 micromoles of adenine to blood stored in ACD pH 5.3 and inosine did not significantly alter the level of 2,3-DPG or P50. (Author)
