A Fluorometric Assay of Methyl Ketones.

Abstract

A quantitative kinetic fluorescence method is reported for alpha-methyl ketones, e.g., acetone and acetophenone. The reaction involves the condensation of o-nitrobenzaldehyde with the alpha-methyl ketones at pH 12.5 to form the fluorescent indoxyl. The rate of fluorescence formation was shown to be linear with respect to methyl ketone at low concentrations. The rate limiting step in the reaction is the one involving the alpha-methyl ketone anion attack on the o-nitrobenzaldehyde. Photolysis of the adduct of the methyl ketone and o-nitrobenzaldehyde likewise yielded indoxyl. Isopropanol by prior oxidation to the ketone was detected also. (Author)
