Isolation of Infectious RNA from Certain Arboviruses of Groups A and B (Vydelenie Infektsionnoi RNK iz Nekotorykh Arbovirusov Grupp A i B),
Abstract
The infectious RNA may be isolated with cold phenol from Japanese encephalitis virus. Treatment with hot phenol is necessary for isolation of RNA of Venezuelan equine encephalomyelitis virus. Removal of residual phenol with ether and nitrogen increases the yield of infectious RNA approximately 10-fold as compared with alcohol reprecipitation. A considerable increase in RNA titers results from dilution of preparations for inoculation of cell cultures in tris buffer with 1 M NaCl solution, pH 7.8 and washing of cultures with hypertonic solution before inoculation. The yield of VEE virus RNA was up to 0.25% of infectivity of the original virus. (Author)
