Development of an Implantable Sustained Release System for the Prevention of Malaria.
Abstract
Polymers and copolymers were synthesized from lactic and glycolic acids. Long term release of antimalarial chemicals from these polymers, based on careful in vitro testing was carried out to evaluate the potential as sustained release implants. Sustained release of WR 142490 over a period of 21 weeks was obtained at levels of 0.01 and 0.04 mg/day sq cm surface area using a higher molecular weight bulk polymerized polylactic acid. Release rates of WR 142490 and polymer hydrolysis rates for four sets of polymer/chemical matrices were obtained. Both bulk and solution polymerized polymer, each at different reaction conditions, were used in these studies. Over-all the release rate of the chemical and the polymer hydrolysis were both shown to be more strongly dependent on chemical loading of the polymer/chemical matrix than on the type of polymer used in these tests.
