Characterization of Cellulase from the Abscission Zone of Bean Plant.

Abstract

Cellulase (beta-1,4-glucan-glucanohydrolase) extracted from bean explants was separated into multiple molecular forms. Cellulase activity was observed at three isoelectric points, 3.5, 7.0, and 9.0. The enzymes was separated into two peaks of cellulase activity using gel filtration and two major peaks and one minor peak using ion-exchange chromatography. Three molecular forms of cellulase appear to be present in bean explants: a major form at pI 9, a second major form at pI 3.5, and a minor form at pI7. The two major peaks separated on G-100 Sephadex have molecular weights of 66,000 and 45,000, respectively, and are believed to be most active in cell separation. (Author)
