Electrophoretic Screening Procedure for Detection of Heme-Variable Hemoglobins.

Abstract

Certain abnormal hemoglobin variants contain less than the normal number of heme moieties. These variants are unstable and are associated with hemolytic anemias and reduced oxygen carrying capacity. Various approaches for detecting heme-variable hemoglobins are described. These include electromagnetophoresis which combines the effects of an electric field on the hemoglobin ionization state with the effects of a magnetic field on the paramagnetic susceptibility of the heme groups; electrophoresis coupled with the spectrophotometric measurement of absorbances at 278 nm and 415 nm; and electrophoresis coupled with dual straining using protein-specific and heme-specific chromagens. The dual staining approach appears to be the method of choice with respect to reproducibility, ease of operation, and long term reliability. (Modified author abstract)
