The Detectability of the NADH Fluorescence Signal.
Abstract
Approaches to the detectability of NADH fluorescence in isolated organelles (mitochondria), intact cells, and in organs (heart, lung, liver, brain) has required a detailed study of components ranging from excitation light source, optical geometries, detectors, and appropriate blocking filters. The use of laser techniques to induce metabolic transients in photosynthetic systems or CO-inhibited respiratory systems in the presence of oxygen together with the spectrophotometric and fluorometric read out of optical signals has further been explored. An evaluation of these sytems in mitochondria, in perfused organs (heart and liver), in the brain, and in the skin of intact animals have been achieved as have new phases of ultra-rapid reactions using laser photolysis in photosynthetic and respiratory systems. Future applications to a variety of biomedical problems were prognosticated. (Author)
