Selective Concentration of Bacteria and Rickettsia with the Aid of Polycondensation Immunosorbents,

Abstract

The authors suggest a method of express analysis for detection and identification of pathogenic microorganisms. The method includes 3 stages: preparation of specific immunosorbents by acting upon globulins of corresponding hyperimmune sera with 4,4-bisdiphenylendiasonium; conjugation of immunosorbents obtained with microorganisms; preparation of smears from the precipitates; the smears are then treated with specific fluorescent serum and studied microscopically under fluorescent microscope. There were established the optimal conditions of polycondensation reaction in obtaining immunosorbents from globulins of hyperimmune sera of P. pestis and D. sibiricus rickettsia. A possibility was demonstrated of lyophilization of immunosorbents without any loss of their activity. With the aid of the suggested method of concentration of microorganisms it was possible to increase the sensitivity of detection of the required microorganisms 10-15 times in comparison with the classic immunofluorescence method.
