Comparison of different Variants of the Colorimetric Method for Determining RNA and DNA in the Mycelium of 'Pen. nigricans Thom',
Abstract
Colorimetric methods for determining DNA with indole and with diphenylamine in modifications show good coincidence of results both in an extract obtained from the mycelium of Penicillium nigricans by the Schneider method and also in an extract by the Schmidt-Thannhauser method. During determination of RNA with orcinol a brownish green tint is formed. The brown tone is eliminated by extraction with chloroform with simultaneous reduction in the optical density of the solution. Precipitation with ethanol from extracts obtained by the Schmidt-Thannhauser and the Schneider methods removed the substance with a polysaccharide nature. In this case there was a small reduction in values for DNA and an essential reduction for RNA. However, the differences in data from colorimetric determinations of RNA in extracts obtained by different methods were not eliminated.
