Sorption of Lecithinase 'C' by Ion-Exchange Celluloses,

Abstract

The object of the work was to study the sorption of lecithinase C from filtrates of a Clostridium perfrigens culture by anion-exchange celluloses. The lecithinase C was sorbed under dynamic conditions on cellulose ionites from filtrates of the culture, concentrated by precipitation with sodium hexamethaphosphate at a pH of 4.2 in the presence of 15% NaCl. Besides protein components, the concentrate contained substantial quantities of nucleic and pigment impurities. Sorption of lecithinase C from culture filtrates improves with an increase in the exhange capacity of ECTEOLA cellulose and the basicity of the ionite; sorption decreases with an increase in NaCl concentration. A simple process was developed for the separation and purification of lecithinase C by successive chromatography on ECTEOLA and DEAE celluloses.
