Utilization of Gas Sparger for Submerged Culture Growth of L Cells and Manifold for Transfer Operations.

Abstract

A gas sparger previously described for use in Bellco culture flasks has been tested in the laboratory to grow L-DR substrain mouse cells in a chemically defined medium. Adequate and continued propagation of L cells has been accomplished by using the criteria of doubling time, final population density, and recovery analysis following each 4-day passage. An inoculum of 5.24 x 1,00000.0 cells per milliliter (19.1 log (base 2) units) grew to a peak population level of 3.3 x 1,000,000.0 cells per milliliter (21.7 log (base 2 units) at the end of the log phase. A population doubling time of 25 hours was observed in the growth cycle of L-DR cells. In addition, the study showed that one ml of packed cell volume was equivalent to 5.46 x 1,000,000.0 cells. Extension of this value led to a determination of the productive output of the suspension gas culture system over a period of 32 days.
