PHOSPHOLIPID COMPONENTS OF SINDBIS VIRUS,

Abstract

Sindbis virus, labeled with p32 during propagation in chick fibroblast monolayers and purified by differential and sucrose density gradient centrifugation, was extracted for lipid with chloroform and methanol (2:1) at 50 C. The p32-labeled lipid extract was examined for phospholipids by thin- layer and paper chromatography. Phosphatides were located by radioautography and identified by comparison with nonlabeled standards and known mobility values. In addition to sphingomyelin, phosphatidyl choline, and phosphatidyl ethanolamine, other phospholipids were tentatively identified as phosphatidyl inositol, phosphatidyl serine, phosphatidyl glycerol, and diphosphatidyl glycerol. Gas chromatographic analysis of the total fatty acids from purified Sindbis virus and normal or infected chick fibroblast cells indicated that palmitic, stearic, and oleic acids were major fatty acid components. In addition, there appears to be more short-chain fatty acids (C-8 to C-16) and fewer long-chain fatty acids (C-20 and above) in the viral lipid than in the normal or infected chick fibroblast cell lipid.
