GROWTH OF RIFT VALLEY FEVER VIRUS IN MONOLAYER CULTURE,

Abstract

Optimum adsorption of Rift Valley fever (RVF) virus to L cells in monolayer culture occurs between pH 6.6 and 7.0. Optimum virus growth occurs between pH 7.6 and 8.0. The virus yield per cell under these conditions is about 1000 MICLD50. The growth of RVF virus in monolayer cultures, infected with a multiplicity designed to infect all the cells at one time, is characterized by a latent phase of about 6 hours, a period of accelerated virus growth resulting in peak virus titer of both cell-free and cell-associated virus between 16 and 20 hours, and a phase of constant virus titer that lasts about 48 hours after peak titer is attained. Cytopathic effects of infected cells developed slowly; they were obvious by 15 hours and marked by 24 hours postinfection. The virus appears to be released continuously form infected cells rather than in a single burst. (Author)
