FAILURE TO FIND AN ACTIVATED ARGININE RESIDUE IN THE ACTIVE SITE OF CHYMOTRYPSIN.

Abstract

This report describes an attempt to confirm the presence of an activated arginine residue in an active-site peptide isolated from an enzymic hydrolyzate of isopropyl methylphosphonofluoridate (GB)-inactivated chymotrypsin (IMeP-chymotrypsin) reported by a previous investigator. The present results failed to confirm his finding. Arginine was not found in the peptide either before or after treatment with ammonia. The composition of the peptide was found to be Gly.Asp.IMePSer.Gly.Gly.Pro.Leu. An unknown, basic, ninhydrin-reacting substance was detected after ammonia treatment of the peptide. (Author)
