EFFECT OF CHEMICAL MODIFICATIONS ON THE BIOLOGICAL ACTIVITY OF TRANSFER RNA,
Abstract
An investigation was made of the effect of chemical modification of s-RNA on its biological activity. In particular a halogenation reaction - (bromination) - and an alkylation reaction - (methylation) - were studied. This work was preceeded by a preliminary chemical and physicochemical study, carried out on nucleotide bases and ribonucleic acids. In addition to these chemical modifications, made on isolated yeast s-RNA, a modification was made of s-RNA by physiological means in adding 5-fluorouracil to the culture medium of the yeast and by isolating the s-RNA, in which 5-FU has been incorporated instead of uracil. The following effect of these chemical modifications on the various biological activities of s-RNA were examined: (1) Its capacity to bind and accept aminoacids; (2) its capacity to transfer the aminoacids; (3) its ability to reattach the terminal CCA triplet; (4) its ability to attach itself to the ribosomes.
