LIPIDS OF VENEZUELAN EQUINE ENCEPHALOMYELITIS VIRUS AND THEIR RELATIONSHIP TO THE LIPIDS OF THE HOST,

Abstract

Venezuelan equine encephalomyelitis (VEE) virus derived from chick embryo and chick fibroblast (CF) cells was comprehensively analyzed for fatty acids, phospholipids and neutral lipids, to determine whether differences in the lipid composition of the virus propagated in these host systems could be related to differences in the lipid composition of the two hosts. The major fatty acids present in the phospholipid fraction of chick embryo - derived VEE virus were palmitic (C16), stearic (C18), and oleic (C18:1) acids. These acids comprised approximately 55% of the phospholipid fatty acid content of the chick embryo host as well as of the virus. The predominant fatty acid was palmitic acid (23 to 27%). Phospholipid fractions of the CF host and CF-derived virus contained the same three major fatty acids; however the predominant fatty acid in these fractions was stearic (21 to 23%). The phospholipid composition of the chick embryo - derived virus was similar, but not identical, to the phospholipid composition of virus derived from the CF host. However, both the embryo-derived and CF-derived virus phospholipid fractions differed quantitatively from the phospholipid fractions of their respective hosts. Phosphatidylcholine was the main phospholipid component (42 to 48% in both types of virus). Thin-layer chromatograms of the chick embryo - derived and CF-derived virus neutral lipids indicated that only two main components were present. These were tentatively identified as cholesterol and diglycerides. In addition to these two components, the neutral lipid fractions of the two hosts contained at least three other components not found in the virus neutral lipid fraction. (Author)
