Photoacoustic and Ultrasonic Image-Guided Needle Biopsy of the Prostate

Abstract

We have optimized interstitial light sources to yield multi-spectral photoacoustic (PA) imaging data in excised prostate tissue. Two types of interstitial sources a directional source and an omnidirectional source have been optimized and validated in gelatin (tissue mimicking)and tissue phantoms with exogenous and endogenous contract. Imaging with an external linear and transrectal array has been demonstrated in real-time at tissue depths in excess of 7 cm and with improved spectral fidelity (i.e., reduced redshift in obtained spectra) using the Verasonics ultrasound platform, is suited for eventual clinical trials. The Verasonics acquisition is performed in a Delrin holder and coordinated with a LabVIEW data acquisition device to allow for energy correction and eventual 3D spatial registration. Model-based reconstruction of the oxygen saturation distribution (and underlying fluence and PA pressure distributions) has been developed. This preliminary work has laid the foundation for prostate biopsies augmented with interstitially driven PA imaging.
