89Zr Trastuzumab PET, Rapid Autopsies, and Patient Derived Xenografts to Determine the Extent of Clonal Evolution in Treatment RefractoryHER2 Gastric CancerDetermine

Abstract

The short-term milestones of this project include; 1) complete accrual to Phase II afatinib/trastuzumab trial and89Zrtrastuzumab PET trial, 2) establish a tumor and PDX bank from 8-10 autopsies, representing the diversity of gastric metastasis; 3) characterize genetic features of tumor clonal evolution, using DNA, RNAseq and miRNA analysis; 4) explore functional significance of the observed alterations in established PDX models; and 4) correlate distinct HER2 gastric cancer signatures in order to better understand factors driving tumor progression and drug resistance.
