Mechanisms of Oncogenesis in the Primary Liver Cell Cancer Cholangiocarcinoma

Abstract

During the reporting period significant progress has been made on understanding the regulatory mechanisms of YAP tyrosine phosphorylation as well as the impact of YAP tyrosine phosphorylation on both YAP transcriptional activity, as well asoncogenic fate, and sensitivity to a standard of care chemotherapy. Specifically we have now demonstrated the tyrosine phosphatase SHP2 can directly regulate YAP activity and chemosensitivity through down regulation of YAP tyrosine phosphorylation. These data have just been accepted for publication in Molecular Cancer Research. Furthermore we have now observed that downregulation of YAP tyrosine phosphorylation is associated with a phenotype switch from cholangiocarcinoma to more predominantly hepatocellular carcinoma.
