Bioinspired Acellular Strategies to Optimize the Skin-Implant Interface of Osseointegrated Devices

Abstract

This work directly addresses PRORP-Peer Reviewed Orthopaedic Research Program 2019, Applied Research Award; Focus Area: "Skin-implant interface-identification of best practices to address infection at the skin-implant interface for osseointegrated prosthetic limbs." We also address residual limb skin integrity and durability. We address the general issue of prevention of infections and skin-to-implant healing and attachment for osseointegrated (OI) prostheses. Our reasoning centers on preventing OI prostheses infection by a simultaneous 2-tier protection system for improving skin healing and adhesion to the OI implant as it exits the skin and improving durability of this seal for implant longevity by controlling biofilm and microbiome challenge. We will engineer multifunctional, acellular materials deriving inspiration from (1) existing biological settings where a hard tissue or organ exits the body through the skin and the biomolecular/cellular structures that seal the attachment of the soft tissue on the hard tissue and (2) natural biomolecules that provide defense against infection.
