Targeting the Cell Surfaceome of Aggressive Neuroendocrine Prostate Cancer
Abstract
In this reporting period, we completed and published studies related to the therapeutic-targeting of CEACAM5 expression in neuroendocrine prostate cancer (NEPC) using the CEACAM5 antibody-drug conjugate labetuzumab govitecan. We have also performed additional characterization of in vitro neuroendocrine trans-differentiation models using defined genetic perturbation of prostate adenocarcinoma cell lines. These analyses indicate that these models recapitulate critical aspects of the human disease, provide a way to rapidly derive molecular insights into the pathogenesis of NEPC, and may be useful to recapitulate the heterogeneity of human prostate cancer. Lastly, we have performed cell-based binding assays to characterize the affinity and specificity of six fully humanized L1CAM CE7 candidate antibodies. We intend to use these in upcoming studies to evaluate their therapeutic potential in NEPC.
