The Role of an Aggrecan 32mer Fragment in Post-Traumatic Osteoarthritis
Abstract
Overall, we sought to develop a neutralizing antibody against the 32mer aggrecan fragment. However, we determined during the course of the project that the antibody failed to block the action of the 32mer in vitro, which prevented further in vivo development. Concurrently, we also sought to develop an assay to measure the aggrecan 32mer fragment in serum, because we discovered that the32mer has pro-algesic activity in experimental OA. We successfully measured the 32mer in both mouse and human serum, and provided evidence that levels were elevated in experimental osteoarthritis. The 32mer has great potential as a novel biomarker for OA pain, as well as a target marker for aggrecanase activity.
