Screening for and Preclinical Evaluation of Drugs for Hearing Loss

Abstract

Screen new small chemical libraries for neurite elongation promoting effects in culture; Elevate hits from the screen to evaluate in an animal model of noise induced hearing loss (NIHL); investigate mechanism of fluvastatin promotion of neurite elongation in culture using chemical inhibitor of the most likely effectors of the pathway; Determine whether fluvastatin stimulates neurite elongation invo after depletion of hair cells; investigate mechanism of fluvastatin protection of hearing synapses and nerve fibers in vivo using geranylgeraniol and H1152; evaluate new modes of statin delivery for hearing protection. This focus changed a bit when we discovered that statins delivered to noise exposed cochleas protected against noise induced hearing loss. We started evaluating statins and hearing and cochlear morphology and focused more on invivostudies.
