Before the Chicken or the Egg: Modulating Ovarian Hormonal and Inflammatory Inputs to Interrupt Carcinogenesis in a Novel Murine Model of Spontaneous EOC

Abstract

We have previously identified a signal of EOC reduction and modulation of inflammatory signaling with MCI-030 potentially in the context of progesterone (P4) signaling. While tantalizing, identification of pre-malignant lesions in the hen model has proven difficult, preventing us from firmly concluding on the effectiveness of intervention. We aim to validate the anti-carcinogenic activity of MCI-030 in a mammalian model that allows visualization at the single cell level of its impact. Given evolving data that P4 acts to clear p53 mutant clones by activating necroptosis we propose a 4-arm study in the novel MADM mouse model to assess the role of PDE10 in progesterone signaling and the impact of inhibition with MCI-030. This model will simultaneously allow for measurement of chemoprevention efficacy with an output of pre-malignant clonal outgrowth.
