Biological Properties of Dry Pertussis Allergen and Methods of Its Standardization,

Abstract

Data obtained by the authors propose that a dry preparation of pertussis allergen use the following indices for standardization. One cutaneous dose of preparation should contain seven micrograms of protein for children and 14 micrograms of protein for guinea pigs with a tolerance of plus or minus 1 microgram. The intracutaneous introduction of one cutaneous dose of allergen to a guinea pig weighing 450-500 g, which had been immunized with pertussis vaccine, should cause in 20-24 hours a skin reaction in the form of an infiltrate and redness 15-20 mm in diameter. In control animals there should be no positive reactions to the introduction of allergen. Reactions are possible in the form of infiltrate and redness up to 5-7 mm in diameter. During the intraperitoneal introduction of 2.5-5 cutaneous doses to white mice weighing 14-16 g, the allergen should not cause the death of the animals. The minimum precipitating dose of pertussis allergen in the ring precipitation test with immune rabbit serum (titer of agglutinins 1:51,200) should not exceed 1/16 of the cutaneous dose.
