Gene Expression in Abnormal Cell Types of 'Neurospora crassa'.

Abstract

Significant results obtained during the first two years were as follows: (1) Abnormal morphology of the 'slime' morphological mutant of N. crassa was correlated with possible changes in DNA composition. (2) The transcribed DNA sequences if can be isolated, proved to be useful indicator for identification of transcriptional differences among mutant strains of N. crassa. (3) A marked differences in transcriptional differences in development of N. crassa was noticed and measured quantitatively. (4) All of the fungal species tested contained 10-20% repeated DNA sequences having 100-110 copies of each sequences. (5) Evidences were obtained on the existence of a new way of flow of genetic information i.e. presence of RNA dependent DNA polymerase (RDDP) activity. (6) Further studies with RDDP enzymes in N. crassa indicate that RNA templates used by RDDP in N. crassa was 6s and not 70s which is typical for oncogenic viruses. This enzyme activity was always found to be bound to RNA, was always RNase sensitive, and requires all four nucleotides for maximum reaction.
