Analogs of Cordyceptin and Adenine Arabinoside (ARA-A) as Potential Antimalarial Nucleosides.

Abstract

The work described in this report deals with the synthesis of 3'-C-alkyl analogues of the nucleoside antibiotic cordycepin as potential antimalarial agents with enhanced liphophilicity, improved membrane transport properties and resistance to enzymatic deamination. In part the work was concerned with the synthesis of lipophilic ester and amide derivatives of Ara-A 3',5'-cyclic phosphate which might show enhanced transport into cells by passive diffusion. Thirty-three compounds were submitted to WRAIR for antimalarial assay, of which four were target cordycepin analogs of the 9-(3'-alkyl-3'-deoxyribofuranosyl)adenine type and one was a 9-(3'-alkyl-3'-deoxyxylofuranosyl)adenine.
