Development of an On-Line Biological Detector
Abstract
Mammalian cells are being grown in artificial capillary perfusion culture in an effort to develop a sensor for toxic components in recycled water intended for human consumption. Cultures of BHK or L929 cells can be established in capillary culture in one week or less by planting the units with a high seed population. Culture using either 1% or 10% calif serum-containing medium allows the cells to reach a plateau in growth. Cultures can be maintained in either a once-through perfusion mode or a recirculation perfusion mode for prolonged periods at rates slow enough to allow rapid measurement of oxygen and glucose utilization and lactic acid excretion rate provided that (1) the flow rate is slow enough and (2) the toxin is in direct contact with the cells.
