Chemotherapy of Rodent Malaria: Evaluation of Drug Action against Normal and Resistant Strains, Including Exo-Erythrocytic Stages.
Abstract
Blood schizontocidal action of 10 WRAIR compounds and tissue schizontocidal action of 75 compounds is summarised in this report. Five-substituted 8-aminoquinolines show particular promise having high level activity of both types, including against chloroquine resistant lines. Residual activity is a feature of this type of structure. In tests against the mefloquine-resistant line of P. berghei WR 225,448 has shown good activity. A screening system is being developed in conjunction with WHO for the detection of blood and tissue schizontocides with activity of long duration. Twenty WRAIR compounds are currently under study in this system. Preliminary data are provided. Several compounds show good residual activity by the oral as well as subcutaneous routes. Investigations are also being carried out on the duration of action of pyrimethamine and other compounds in nondegradable and biodegradable implants. The Alzet minipump has also been used for this study. Further investigations are reported on the mode of drug action of chloroquine and mefloquine. Screening activities are currently focusing on the evaluation of delivery systems for tissue schizontocides to produce are a radical cure of vivax malaria. (Author)
