Studies on PGBx A Polymeric Derivative of Prostaglandin B1. I. Synthesis and Purification of PGBx.

Abstract

PGB sub x, a new polymeric derivative of PGB sub 1, previously was shown to restore oxidative phosphorylation to degraded isolated rat liver mitochondria in vitro, and to reverse the effects of cardiogenic ischemia in monkeys and cerebral ischemia in rabbits. This report describes in detail the synthesis and purification of PGB sub x via PGB sub 1, starting with azelaic acid. In addition, details of the in vitro mitochondrial assay are reported. Purified PGB sub x exhibiting maximal reactivation of mitochondrial phosphorylation has a mean molecular weight of 2350. The yield of PGB sub x based on azelaic acid was 4% and based on PGB sub 1 was 25%. (Author)
