Mode of Action of Polyamine Analogues on the Growth and Biochemistry of Leishmanial Cells.

Abstract

In the present study we confirmed the occurrence of polyamines in leishmanial promastigotes and showed that their synthesis is catalyzed by ornithine decarboxylase. The activity of this enzyme fluctuates during the growth cycle of leishmanial promastigotes and amastigotes. Dramatic changes in cellular polyamine levels and in the activity of ornithine decarboxylase were observed in macrophages infected with leishmanial amastigotes. It was shown that sinefungin, an ornithine-nucleoside, possesses potent antileishmanial activity and inhibits the growth of both leishmanial promastigotes and amastigotes. New aminoalkyl-anthraquinone derivatives have been synthesized; some of these exhibit potent antileishmanial activity. Compound WR-6026 was tested for its antileishmanial activity.
