Drug Development Against Viral Diseases of Military Importance.

Abstract

This report discusses the development of routes to analogs of carbocyclic 3-deazaadenosine that are incapable of phosphorylation at the 5'-position and acyclic analogs of pyrazofurin. Functional group manipulation at C-5' of the nucleoside analog has allowed the preparation of 5'-amino-5'-deoxy and 5'-phthalimido-5'-deoxy derivatives. Progress toward other targets with halo and thio groups at C-5' as well as targets with C-5' oxidized is reported. Initial work heading toward a suitable pyrazole derivative for attachment of various side chains in the synthesis of acyclic pyrazofurin analogs is also described.
